Item Cholesterol lowering effects of bovine serum immunoglobulin in human participants with mild hypercholesterolemia(Texas A&M University, 2006-10-30) Black, Melinda LoriHypercholesterolemia is a major risk factor for cardiovascular disease (CVD). Interestingly, the consumption of dairy products, namely milk, has been shown to lower cholesterol. The mechanism of action surrounding this observation has been attributed to the protein fraction of milk. While there have been many studies evaluating the effects of dietary protein sources on cholesterol concentrations, few studies have evaluated specific animal protein components and no human clinical studies regarding the effects of animal plasma protein fractions on cholesterol metabolism have been conducted. This study examined the effect of an oral serum bovine immunoglobulin protein fraction (bIg) derived from US Department of Agriculture approved beef (aged < 30 months) on lipid indices in hypercholesterolemic humans. Participants included men and women (aged 25 ?????? 70 years) with mild hypercholesterolemia (5.44-6.99 mmol/L) who were not receiving cholesterol-lowering medication. Treatment consisted of the randomized, double blind, parallel group, placebo-controlled administration of 5 grams (g) bIg daily for 6 weeks (W) in 52 participants (n = 26 each in treatment and control groups). Mean (???? SD) baseline treatment and placebo total cholesterol (TC) was 6.33 ???? 0.1 mmol/L and 6.16 ???? 0.1 mmol/L respectively. A repeated-measures multivariate analysis of covariance (MANCOVA) covaried for change in total energy and alcohol intake, and a Tukey posthoc examination of the data showed that the bIg-treated group demonstrated a significant reduction in TC at 3-week (W) (5.98 ???? 0.5 mmol/L; P < 0.05) and 6-week (W) (5.97 ???? 0.7 mmol/L; P > 0.05) intervals compared to baseline. The 6W concentration was significantly lower than the placebo group (P < 0.05). Additionally, study findings displayed no significant changes in the placebo group or in any other lipid indexes or markers associated with hepatorenal or cardiovascular health. Consumption of bIg appears to lower major lipid indexes associated with CVD.Item Dietary polyunsaturated fatty acids modify plasma lipids and red blood cell membrane composition but do not induce ∆6 desaturase mediated conversions in the domestic feline(2009-05-15) McClure, Melena KathleenThis study explored the effects of dietary unsaturated fatty acids on feline lipid metabolism. It was hypothesized that high dietary linoleic acid (18:2n-6, LA) would enhance conversion to arachidonic acid (20:4n-6), enrichment of dietary long chain n-3 FA (LCn-3FA) would affect lipid parameters, and n-3 FAs incorporation may blunt n-6 FA incorporation. Twenty-nine cats were randomized into groups (n = 9, 10, 10), and fed for 28 days with blood collections on days 0, 14, and 28. Experimental diets consisted of a commercial diet, supplemented with 8g oil/100g kibble. Oil supplements and subsequent diets were: high-oleic sunflower (H diet) with 82% oleic acid (18:1n-9), Menhaden fish (M diet) with LCn-3FA, and safflower (S diet) with 75% 18:2n-6. Dietary 20:4n-6 content was: 0.03 for H and S, and 0.09 for M (g FA/kg diet). Nonesterified fatty acid (NEFA), triacylglycerol (TG), total cholesterol (TC), lipoproteincholesterol (LP-C), plasma phospholipid (PL) FAs, red blood cell membrane (RBC) FAs, and ?5 and ?6 desaturase indices were measured. Statistical analyses were performed with SAS PROC MIXED with p < 0.05 determining significance. Neither TC nor NEFA showed significant effects. Diet M resulted in significant TG lowering, despite typically low feline TGs. Similarly, pre-? LP-C (i.e. TG-rich VLDL) was decreased in diet M. Plasma PL FAs revealed significant accumulations of the following: 18:1n-9 in diet H, 18:2n-6 in diet S, and LCn-3FA in diet M. Despite high dietary 18:2n-6, plasma PL 20:4n-6 was not increased in diet S over diets H or M. Increased docosadienoic acid (20:2n-6) in diet S demonstrated that 18:2n-6 chain elongation occurred in deference to its ?6 desaturation further substantiating low feline ?6 desaturase activity. Interestingly, no diet M blunting of 20:4n-6 incorporation occurred because fish oil supplementation provided additional 20:4n-6. Tissue 20:4n-6 content appears to be diet-dependent. Accumulation of eicosapentaenoic acid (20:5n-3), but low affinity for docosahexaenoic acid (22:6n-3) occurred in diet M RBC membranes. After 28 days, plasma PLs reflect dietary intake more readily than RBC membranes. Fish oil supplementation resulted in plasma PL LCn-3FA enrichment and lowered plasma TG concentrations, both of which may have physiological significance in cats.Item Effect of dietary polyunsaturated fatty acids and related nutrients on sebum lipids, and skin and hair coat condition in canines(Texas A&M University, 2005-02-17) Kirby, Naomi AnneA study was performed to investigate the effect of diets rich in polyunsaturated fatty acids and other related nutrients, in the effort to improve skin and hair coat conditions in canines. The study included 24 dogs fed a baseline diet (Ol?Roy?), with an acclimation period of 12 weeks (Phase I). Nine female beagles and 15 male hound mixed-bred dogs were used. For the next 12 weeks (Phase II) the dogs were randomly divided into three groups and fed one of three specially formulated complete and balanced diets. Differences among the three diets were as follows: Diet A contained lower but adequate amounts of dietary zinc and linoleic acid than Diet B. Diet C was similar to Diet B with respect to zinc but contained less linoleic acid and more alpha-linolenic acid. In the beginning, a preliminary study assessing skin lipids showed that hair is a suitable material to analyze. This study lead to the subsequent investigation, using plucked and shed hair samples obtained on weeks 3, 7, and 11 (Phase I) and again on weeks 1, 3, 7, and 11 (Phase II). One objective of this research was to investigate the dietary effects of the combination of linoleic acid in combination with zinc on canine sebum lipids during a 12 week diet phase. Another objective was to investigate if any correlation existed between the hair cycle anagen and telogen phase of the hair follicle and the lipid constituents present in the sebum during the 12 week acclimation phase and the 12 week feeding trial. Two hypotheses were tested. First, the lipid content of hair can be altered by fat and fatty acid composition of the diet. Second, the modification of dietary fat and polyunsaturated fatty acids lead to changes in hair lipids that may be related to improved skin and hair coat scores. Over the 12 week feeding period of Phase II, test results revealed statistically significant increases in both CE and CE/WD, and a prolonged growth of hair follicles in the anagen phase of diet B dogs, which are both consistent with improved skin and hair coat scores.
